ABSTRACT

Objective To investigate the distribution of mosquito species and their associated viruses, and identify Culex pipiens subspecies in Hami City, Xinjiang Uygur Autonomous Region. Methods Mosquitoes were captured using mosquito trapping lamps method in Yizhou District, Yiwu County, and Balikun County of Hami City in mi-July, 2019 and 2020. The species and subspecies of all captured mosquitoes were characterized. In addition, the flavivirus, alphavirus, bunyavirus, Japanese encephalitis virus, Liaoning virus, Tahyna virus, tick-borne encephalitis virus and West Nile virus were detected using reverse-transcription PCR assay in captured mosquitoes. Results A total of 1 496 mosquitoes were captured from Yizhou District, Yiwu County, and Balikun County of Hami City, belonging to 3 genus and 3 species. Cx. pipiens was the dominant mosquito species (986 mosquitoes, 65.91%), followed by Aedes caspius (457 mosquitoes, 30.55%), while Culiseta alaskaensis had the lowest number (53 mosquitoes, 3.54%). All captured Cx. pipiens mosquitoes were identified as Cx. pipiens pipiens based on the terminalia of male mosquitoes. RT-PCR assay tested negative for flavivirus, alphavirus, bunyavirus, Japanese encephalitis virus, Liaoning virus, Tahyna virus, tick-borne encephalitis or West Nile virus in captured Cx. pipiens mosquitoes. Conclusions There were 3 species of mosquitoes in Hami City from 2019 to 2020, including Cx. pipiens, Ae. Caspius and C. alaskaensis, with Cx. pipiens as the dominant mosquito species, and all captured Cx. pipiens mosquitoes were Cx. pipiens pipiens; however, no arboviruses were detected.

ABSTRACT

OBJECTIVE@#To evaluate the effect of femoral and sciatic nerve block in total knee replacement of elderly patients under general anesthesia.@*METHODS@#From July 2017 to July 2019, 60 patients with unilateral total knee replacement were selected, including 35 males and 25 females; aged 66 to 74(70.2±10.3) years;BMI 18 to 25 (21.3 ± 3.5) kg /m;course 2 to 3 (1.2±0.3) days. The patients were divided into general anesthesia group (G group) 30 cases and general anesthesia plus nerve block group(GNB group) 30 cases. In GNB group, the femoral nerve sciatic nerve block was guided by ultrasound before anesthesia induction, 20 to 25 ml was injected into the femoral nerve puncture point with 0.5% ropivacaine, 15 to 20 ml was injected into the sciaticnerve puncture point, and the total volume was no more than 40 ml. Postoperative intravenous analgesia (PCIA) was performed in two groups. The dosage of propofol and remifentanil was recorded. Forty-eight hours after operation, the incidence of postoperative nausea and vomiting (PONV) and postoperative farsightedness were recorded. When VAS>3, tramadol 2 mg / kg was injected intravenously, and the additional times of tramadol were recorded. Forty-eight hours after operation, patients' satisfaction score was used to record the length of stay.@*RESULTS@#Compared with group G, the dosage of propofol and remifentanil decreased, the incidence of PONV and the number of additional tramadol decreased, and the patients' satisfaction increased (0.05). The ROM and HSS scores of two groups after treatment were higher than those before treatment (0.05). The ROM and HSS scores of the GNB group after treatment were higher than those of the G group (<0.05), and the VAS scores were lower than those of the G group (<0.05).@*CONCLUSION@#The application of femoral sciatic nerve block in total knee replacement under general anesthesia in elderly patients has good postoperative analgesic effect, and can reduce the dosage of general anesthesia, reduce PONV, and increase patient satisfaction.

ABSTRACT

OBJECTIVE@#To study the developmental and morphological characteristics of the mandible in patients with impacted mandibular second molar and to predict the possible trend of mandibular development via three-dimensional (3D) measurement and analysis.@*METHODS@#A total of 88 cases of impacted group and 88 cases of control group were screened out. 3D measurements were performed by using Mimics software. A total of 23 landmark points and 17 measurements were determined. The measurements were analyzed by t-test.@*RESULTS@#The mandible length, the space between the first molars, the space between mandibular angles, and the width between the first molars in the impacted group were lower than those in the control group (P<0.05). Moreover, the value of the submandibular angle was higher than that in the control group (P<0.05).@*CONCLUSIONS@#The impacted mandible of patients with mandibular second molar showed lack of sagittal and width development, and the impacted mandibular second molar was a manifestation of its degeneration.

ABSTRACT

Objective To observe the clinical manifestations of POEMS syndrome, and to improve the early diagnosis and prognosis.Methods The clinical data of 61 POEMS syndrome patients were analyzed retrospectively. Results Among the 61 patients,there were 27 males and 34 females with an average age of 47.2 years.The average time from the primary admission to the confirmed diagnosis was 22.5 months.All the patients(100%)had peripheral neuropathy by various degrees,the majority showing the sensory dysfunction of lower limbs.Fifty-two cases (85.2%)were identified with organomegaly by imaging techniques,most presented with the enlargement of spleen and lymph nodes.Fourty-five cases(73.8%)were accompanied with endocrinopathy, reflected by diabetics along with the hypofunction of the pituitary, gonad, thyroid and the adrenal glands.Fifty cases(82.0%)were with positive serum M protein.Twenty-seven cases(44.3%)manifested evident skin changes including hair growth along the limbs and trunk, skin pigmentation and skin keratinization of the feet.In this group of patients, 59 patients mainly use dexamethasone treatment,some patients with horses flange or cyclophosphamide,lenalidomide treatment. The other 2 patients were treated by autologous stem cell transplantation therapy.The clinical outcome of patients with diagnosis time ≤10 months was significantly better than that of patients with diagnosis time 10 to 20 months and≥20 months(all P<0.05).Patients with diagnosis time ≥20 months had the worst clinical outcomes, with the highest death rate(all P<0.05).Conclusions The clinical manifestations of POEMS syndrome are complex, especially the peripheral nerve damage symptoms and merging other organs or systems.M protein detection is needed in order to make clear diagnosis.Early diagnosis and effective treatment for patients can get better prognosis.

ABSTRACT

Objective To investigate the relationship between epidermal growth factor receptor (EGFR) gene mutation and computed tomography (CT) features and clinical features in non-small cell lung cancer (NSCLCs) patients. Methods The clinical data of 187 patients with NSCLCs admitted in our hospital from September 2014 to July 2016 were retrospectively analyzed. All the patients accepted the EGFR mutated gene detection, and they were divided into effective mutation group (n=67) and non-effective mutation group (n=120). The clinical data and lung CT imaging data were complete in the two groups. The univariate and multivariate Logistic regression analysis were used to analyze the differences of imaging and clinical features between the two groups. Results Comparing with the non-mutation group, there were higher proportion of women and lower smoking index in the EGFR effective mutation group. Lesions in the lung tissue showed a clear edge, leaf and burr, and containing ground-glass opacity (GGO) component, usually accompanied by airway bronchogram and pleural indentation, and associated with cancer lymphatic inflammation and lung metastasis in mutation group (all P < 0.05). There were no significant differences in the proportion of necrosis, cavitation, calcification, halo sign and vacuole sign between the two groups (P > 0.05). Logistic regression analysis showed that female, GGO containing composition, burr, air bronchogram, carcinomatous lymphangitis and lesion density showed obvious airway involvement were the predictive risk factors in patients with EGFR effective mutation. Conclusion The EGFR mutation occurs more oftern in female. The reliable predictive signs of CT include GGO composition, burr, airway bronchogram and carcinoid lymphangitis. In critical patients who are not easy to obtain clinical pathology, it has a guiding significance to radiographic assessment for EGFR effective mutation.

ABSTRACT

Objective To synthesize a quantum dot (QD) to recognize glioma cells for imaging and photodynamic therapy. Methods By one-pot aqueous approach, near infrared-emitting CdTe was produced. After detection of its physicochemical characterizations, RGD was conjugated. Emission images were observed with confocal microscopy. To test its toxicity, CdTe-RGD with various concentrations was separately added into U251 and 3T3 cells for incubation in dark circulation. To test its photodynamic effect, U251 and 3T3 cells were then irradiated for 5 ~ 60 min using 632.8 nm laser. Results The QD (Φ = 3.75 nm, PL peak wavelength =700 nm, PLQY=20%) achieved was a spherical crystal with excellent monodispersity. Under confocal microscope , U251 cells were visualized but 3T3 cells not. In dark circulation, the survival rates of both U251 and 3T3 cells were above 85%. After laser irradiation, the survival rate of U251 cells decreased to (37 ± 1.6)%with the increasing of irradiation time and CdTe-RGD concentration. Conclusion With good physicochemical characterization and low toxicity, CdTe-RGD could be applied in biomedical imaging and photodynamic therapy of gliomas.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of advanced oxidation protein products (AOPP) on expressions of stromal cell-derived factor-1alpha (SDF-1alpha) in ECV304 cells and the signal pathway that mediated the effects.</p><p><b>METHODS</b>AOPP-BSA was made from bovine serum albumin (BSA) and sodium hypochlorite. After treated with AOPP-BSA of different concentrations (50, 100, 200 micromol/L), the expressions of SDF-1alpha mRNA in ECV304 cells were measured by reverse transcription-polymerase chain reaction (RT-PCR) and the expressions of SDF-1alpha protein and the levels of phosphorylated extracellular signal-regulated kinase (ERK) in ECV304 cells were analyzed by Western blot. In inhibition test, U0126, the special inhibitor of ERK of different concentrations (0.1, 1, 10 rmol/L) were added into ECV304 cells culture media for 1 hour, then the cells were treated with AOPP-BSA for 24 hours, at last the protein levels in supernatant were detected by enzyme-linked immunosorbent assay (ELISA).</p><p><b>RESULTS</b>AOPP-BSA obviously promoted the expressions of SDF-1alpha mRNA and increased the levels of SDF-1beta protein of ECV304 cells in dose-dependent manner (all P < 0.01), after 15 minutes treated with 200 micromol/L AOPP-BSA, the levels of phosphorylated ERK of ECV304 cells increased significantly (P < 0.01). When the ERK pathway was blocked by U0126, the promoting effects of AOPP-BSA on expressions of SDF-la protein in ECV304 cells were significantly inhibited in dose-dependent manner (P < 0.05).</p><p><b>CONCLUSION</b>AOPP induced the expression of SDF-la of ECV304 cells, ERK signal pathway is an important pathway that mediated the effects.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the phenotype and genotype of CMTX1 patients with episodic transient reversible white matter involvement, and delineate the features of brain MRI in the episode and the possible mechanisms.</p><p><b>METHOD</b>Three Chinese probands and their family members were sequenced in the coding regions of GJB1. With the other 16 reported CMTX1 patients with episodic transient reversible white matter involvement, the clinical feature of the episodic central nervous system symptoms and the genotypes were reviewed.</p><p><b>RESULT</b>Missense mutations in GJB1 were identified in all 3 probands. In 19 patients with transient reversible white matter involvement, the episodes were manifested as weakness of the limbs, dysarthria, and dysphagia, without disturbance of consciousness or seizures. The episodes lasted for 13 hours (10 min-72 hours) with complete remission in all patients; There were multiple episodes in 9 patients. During the episode, brain MRI showed symmetrical high signals in T2 weighted, Flair and DWI images in periventricular white matter, with predominance in posterior region including splenium of corpus callosum. These changes in imaging were most prominent during or within 1 week after the clinical episode.Significant improvements occurred within 1 month, with complete remission within 4-6 months.No specific locations of mutant amino acids in GJB1 protein were found in these patients with episodic transient reversible white matter involvement.</p><p><b>CONCLUSION</b>Episodic transient reversible white matter involvement may present in a small number of patients with CMTX1. Transient edema of oligodendrocytes due to the dysfunction of gap junction may be involved in the pathogenesis. There is no correlation between the location of the mutant amino acids in GJB1 and the occurrence of the episodes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical effects of the Chinese drug Yimusake, used alone or in combination with trazodone hydrochloride, on primary premature ejaculation (PE).</p><p><b>METHODS</b>Sixty-eight primary PE patients were randomized to a control (n=32) and an experimental group (n=36), the former treated with Yimusake 1 tablet (50 mg) pre day, and the latter with 1 tablet of trazodone hydrochloride (50 mg) pre day in addition, both given orally after supper and for 4 weeks, followed by observation of the therapeutic effects.</p><p><b>RESULTS</b>Eighteen cases (56.25%) responded in the control and 25 (69.44% ) in the experimental group, with statistically significant difference between the two groups (P<0.05).</p><p><b>CONCLUSION</b>Yimusake combined with trazodone hydrochloride is highly efficacious for primary PE.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To compare the efficacy differences between acupoint catgut-embedding combined western medicine (ACCWM) and simple western medicine (WM) for treatment of epilepsy of generalized seizures type.</p><p><b>METHODS</b>Fifty-two patients with epilepsy of generalized seizures type were randomly divided into an ACCWM group (n=30) and a WM group (n=22). Dazhui (GV 14), Yaoshu (GV 2), Jiuwei (CV 15), Qihai (CV 6) were selected in the ACCWM group as the main acupoints combined with routine antiepileptic drugs, Valpromide was taken orally, 2.0 g/d for adult, 50 mg/kg x d for child. The WM group was treated with routine antiepileptic drugs. The epilepsy score and the attack frequency one year before the treatment and one year during treatment were observed and the therapeutic effects in the two groups were compared.</p><p><b>RESULTS</b>The epilepsy score and the attack frequency were both decreased in the two groups (P<0.01, P<0.05), and the epilepsy score in the ACCWM group decreased significantly (6.57 +/- 3.29 vs 10.73 +/- 2.54, P<0.05). The total effective rate of 90.0% (27/30) in the ACCWM group was superior to that of 68.2% (15/22) in the WM group (P<0.05).</p><p><b>CONCLUSION</b>The therapeutic effect of ACCWM is superior to that of simple western medicine in treating epilepsy of generalized seizures type. It can improve significantly the attack symptoms so as to enhance life quality of the patients with epilepsy.</p>

ABSTRACT

·AIM:To report a case of entry of antibiotic ointment into the anterior chamber following sutureless clear corneal incisions phacoemulsification.·METHODS:A 62-year-old man had uneventful phaco-emulsification with clear corneal incisions with intraocular lens(IOL) implantation. Postoperative medication included antibiotic/steroid ointment; the eye was firmly patched at the end of the procedure. On the first postoperative day, the patient was seen an oily droplet floating in the anterior chamber. Immediate surgical removal of the ointment without IOL exchange was performed.·RESULTS:During follow-up for six months after irrigation/aspiration of the anterior chamber and removal of the oily droplet, visual acuity and intraocular pressure (IOP) were unchanged and anterior segment was formed and quiet.·CONCLUSION:Ophthalmic ointment may ingress into the anterior chamber through unstable clear corneal incisions. The case highlights the importance of appropriate wound construction and integrity, as well as the risk of tight eye patching following placement of ointment.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the efficacy and safety of intravesical instillation of high-dose pirarubicin during perioperative period to prevent bladder carcinoma recurrence.</p><p><b>METHODS</b>A prospective randomized controlled clinical trial was carried out. A total of 120 patients with bladder transitional cell carcinoma (TNM I-II) underwent partial cystectomy or transurethral resection of bladder tumour were randomly divided into 2 groups, which were chosen from the patients were hospitalized during June 2003 to May 2009. There were 62 patients were in group A and 58 patients were in group B. In group A, intravesical instillation of high-dose pirarubicin during perioperative period was conducted. In group B, intravesical perfusion of pirarubicin was performed regularly after operation. All the patients were followed up in order to record the relapse rates and adverse reaction.</p><p><b>RESULTS</b>No significant difference was found between group A and group B from gender, age, pathological stage and operation methods (P > 0.05). The recurrence rates were 8.1% in group A, and 20.7% in group B. The adverse effect rates of urethrostenosis in group A and group B were 3.2% and 18.9%. The adverse effect rates of cystic stimulation including irritation signs of bladder and hematuria in group A and group B were 4.8% and 27.6%. There were significant differences in recurrence rates and adverse effect rates between the two groups (P > 0.05).</p><p><b>CONCLUSION</b>Intravesical instillation of high-dose pirarubicin during perioperative period is an effective procedure for the prevention of bladder cancer recurrence.</p>

ABSTRACT

<p><b>BACKGROUND</b>NF-kappaB p65 was shown to inhibit transcription of phosphoenolpyruvate carboxykinase (PEPCK), a rate-limiting enzyme in gluconeogenesis in the liver. To understand the mechanism of action of NF-kappaB p65, we investigated the nuclear receptor corepressor in the regulation of PEPCK transcription.</p><p><b>METHODS</b>Rat H4IIE cells, human hepatoma HepG2 cells and human embryo kidney (HEK) 293 cells were used in this study. The transcriptional activity of a rat PEPCK gene promoter (-490/+100) was analyzed in HepG2 cells, a HepG2 super suppressor IkBalpha (ssIkBalpha) stable cell line, and HEK 293 cells. The effects of p65 and ssIkBalpha on a rat PEPCK gene promoter were observed using the PEPCK luciferase reporter system. The interaction of the cAMP-response- element-binding (CREB) protein, histone deacetylase 3 (HDAC3) and silencing mediator for retinoic and thyroid hormone receptors (SMRT) with the PEPCK gene promoter were investigated using the chromatin immunoprecipitation (ChIP) assay. p65 cotransfection and RNAi-mediated gene knockdown were used to determine the corepressor involved in the inhibition of PEPCK by NF-kappaB p65 and the transcriptional regulation of CREB by NF-kappaB p65.</p><p><b>RESULTS</b>NF-kappaB p65 inhibited PEPCK expression and the inhibition was blocked by ssIkBalpha. The inhibitory effect of p65 was completely blocked in a HepG2 stable cell line in which ssIkBalpha was expressed. HDAC3 or SMRT knockdown led to a significant up-regulation of PEPCK reporter activity in the presence of p65 cotransfection. In the ChIP assay the interaction of HDAC3 and SMRT with the PEPCK gene promoter was induced by p65 activation, but the CREB signal was reduced. Transcriptional activity of CREB was inhibited by NF-kappaB p65 cotransfection. The inhibitory effect of NF-kappaB p65 was blocked by HDAC3 RNAi or SMRT RNAi.</p><p><b>CONCLUSIONS</b>The study showed that the inhibition of PEPCK by NF-kappaB p65 was dependent on HDAC3 and SMRT, which form a nuclear corepressor complex for transcriptional inhibition. The transcription factors NF-kappaB p65 and CREB share the same corepressor HDAC3-SMRT, and the corepressor exchange leads to inhibition of PEPCK gene transcription by NF-kappaB p65.</p>

ABSTRACT

<p><b>BACKGROUND</b>Tourniquets used during total knee arthroplasty may lead to many complications. The aim of this study was to determine perioperative blood loss and its clinical relevance in total knee replacement surgery after applying a tourniquet.</p><p><b>METHODS</b>From June 2009 to October 2009, 60 consecutive patients who underwent routine total knee arthroplasty were randomly divided into two groups and were treated with or without a tourniquet (30 patients/group). There were no significant differences in patient baseline characteristics between the two groups. We compared the two groups of patients in terms of intra- and postoperative bleeding, invisible or visible bleeding, and total blood loss.</p><p><b>RESULTS</b>None of the patients showed poor wound healing, lower extremity deep venous thrombosis or other complications. The amount of blood loss during surgery was lower in the tourniquet group than in the control group (P < 0.01). However, postoperative visible bleeding (P < 0.05) and occult bleeding (P < 0.05) were significantly greater in the tourniquet group than in the control group. There was no significant difference in the total amount of blood loss between the two groups (P > 0.05).</p><p><b>CONCLUSIONS</b>Tourniquet can reduce bleeding during total knee replacement surgery, but is associated with greater visible and invisible blood loss.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate mid-term clinical results of microendoscopic discectomy (MED) for the treatment of lumbar disc hermiation.</p><p><b>METHODS</b>In the study, 117 patients were reviewed,including 63 male and 54 female, ranging in age from 24 to 72 years,with an average of 50.6 years. Among the patients, 60 patients had simple lumbar disc herniation, 10 patients had simple lateral crypt stenosis, 32 patients had lumbar disc herniation combined with lateral crypt stenosis, and 15 patients were combined with calcified nucleus pulposus. Two patients had herniation in L3-L4, 56 patients in L4-L5, 48 patients in L5-S1, 11 patients in L4-L5 and L5-S1. The central type of lumbar disc herniation occurred in 22 patients and the lateral type of herniation occurred in other 95 patients. The Protrusion type of herniation occurred in 32 discs, ruptured type in 73 discs, and free type in 12 discs. Ninety-eight patients had lumbar and leg pain in one side, and 19 patients in double sides. MED was used to remove the nucleus and decompress the nerve root canal.</p><p><b>RESULTS</b>All the patients were followed up and the duration ranged from 48 to 84 months,with an average of 5.5 years. According to lumbar and leg pain evaluation criteria from spinal group of Chinese Orthopaedic Association, there were 93 patients got an excellent result, 16 good and 8 poor.</p><p><b>CONCLUSION</b>Appropriate control indications and skillful surgical techniques are the key points to decrease the complication and to improve the curative effect.</p>

ABSTRACT

Objective To verify the determination of iodine in foodstuff by dry ashing As3+-Ce4+catalytic speetrophotometry.Methods The mixture of foodstuff powder and the solution of K2CO3,ZnSO4,KClO3 and NaCl was heated and dried at 105℃ for 3 hours,then heated by a adjustable electric heater for around 0.5 hour,transferred into muffle fumace to eremated at 600℃ for 4 hours.The dissolved ash was measured by As3+-Ce4+catalytic speetrophotometry.The linear range of the calibration and sensitivity were tested;The precision and accuracy for three kinds of iodine in samples of difierent pumpkins were tested:The iodine contents of standard urine samples and the American standard materials were tested as well.Results This testing covers iodine ranged from 4.4 ng to 250 ng.The relevance coefficient of standard curve was from-0.9997 to-0.9993.The pumpkin iodine contents detected were 45.8,145.0,195.6 μg/kg,with constant variables of 4.3%,3.0%and 3.9%respectively.The recovery was 96.8%,97.8%and 97.6%for three kinds of iodine in samples[(47.2±2.6),(71.9 4-3.3),(95.9±2.4)μg/kg].The relative error was-6.5%when the American standard materials were assessed.The relative error were 11.0%.10.7%and 10.7%when the standard urine samples of three kinds were tested.Conclusion This method,easy to be pefformed with better precision and accuracy,is suitable to measure food iodine as well as total iodine in urine.

ABSTRACT

<p><b>OBJECTIVE</b>The experiments were conducted to study specialty of seed germinating, plant growing and developing and cultural practices of A. edgeworthii.</p><p><b>METHOD</b>The germinating and sowing test, growth habitants, photosensitive reaction of seedling from subterraneous seeds and above ground seeds on A. edgeworthii were studied in this experiment.</p><p><b>RESULT AND CONCLUSION</b>The results indicated that (1) The water could not be absorbed by seed from plant above ground; (2) The underground and above ground seeds could normally germinate, grow, blossom and bear fruits, A. edgeworthii sowed in early May, blossomed in early September and matured in the last ten days of October; (3) The seed leaf of seedling grew underground; (4) The underground seed was produced from subterraneous branch stem which developed from node of seed leaf; (5) A. edgeworthii was shade demanding, high-temperature sensible short-day light plant. The flowering could be greatly advantaged by short-day light treatment (12, 10, 8, 6 h). Cultural practices were studied also in this experiment.</p>

ABSTRACT

Objective To construct the eukaryotic expression plasmid of pEGFPC1uPAR gene and explore the effect on the proliferation and invasion ability of Pam 212 cells. Methods The human uPAR cDNA was cloned by PCR, and inserted into the eukaryotic expression plasmid pEGFPC1. After identification of sequencing, the reconstructive plasmid was transformed transiently into Pam 212 cells, then the cell growth and the invasion ability were evaluated. Results The reconstructive plasmid of pEGFPC1uPAR was validated by sequencing. The reconstructive plasmid can promote the growth of Pam 212 cells and enhance the invasion ability. Conclusion The pEGFPC1uPAR plasmid was constructed successfully and uPAR was confirmed to promote the growth and the invasion ability of Pam 212 cells, which lay the foundation for further studies of uPAR in vivo.
